The aim of this thesis was to address challenges in multi-database Real World Evidence (RWE) studies and to compare modern strategies used in these studies. The findings aim to guide researchers on suitable methodologies for conducting multi-database (MDB) studies and to support stakeholders—including regulatory bodies, health technology assessment agencies, and clinicians—in making informed decisions about medicines.

Specific questions addressed included:
•    How do MDB RWE studies report and handle missing data?
•    Are there differences in patient characteristics, outcomes, and exposure-outcome effect estimates when using a common protocol-only approach, a common data model with study-specific data, or a general CDM?
•    How can heterogeneity in MDB RWE study results inform us about international differences in the use and effects of drugs, as well as the underlying population and healthcare system?
 

The research was conducted at the Division of Pharmacoepidemiology and Clinical Pharmacology at the Utrecht Institute for Pharmaceutical Sciences (UIPS), Utrecht University.
