To assure access to healthcare and treatment in the future, sustainable use of expensive drugs is very important. This research aims to optimize treatment with regard to efficacy and safety, while aiming to reduce drug costs.

The aim of this project is to achieve more effective patient treatments with antibiotics by quantifying the concentrations at the site of infection with minimal sample volumes. To enable quantification of antibiotics at the site of infection, multiple assays should be developed and validated in these infected matrices, such as synovial fluid, synovial tissue, bone, and vitreous humor. Furthermore, the focus will be on small-volume samples and more patient friendly, and sustainable sample preparations.

This thesis explores how systemic inflammation and continuous renal replacement therapy (CRRT) influence drug pharmacokinetics in critically ill patients. Key research questions include: How does inflammation alter midazolam metabolism in critically ill COVID-19 patients? What impact does CRRT have on the drug CL and efficacy of commonly administered drugs at the ICU? The findings aim to optimize therapeutic approaches in critically ill patients.
